The protein chemical analysis of complex I was performed by using mass spectrometry. The subunit composition of complex I and its subcomplexes was analyzed by 2-D gel electrophoresis. Protein spots were identified by MS techniques and three novel subunits have been discovered. Solvents, such as mixtures of cholroform and methanol have enables proteins to be solubilized, often selectively, purified in the absence of detergents and analyzed by ESI-MS.
